Simultaneous Determination of Amlodipine and Olmesartan Using HPLC with Fluorescence Detection

A sensitive, simple and rapid RP-HPLC method with fluorescence detection has been developed to simultaneously quantify amlodipine besylate (AML) olmesartan medoxomil (OLM) in their combined pharmaceutical preparation. The development was divided into three phases. In the first phase, a Plackett-Burman design (PBD) used screen five independent factors. objective of second phase optimize performance using single factor obtain best chromatographic for short run time. Optimum separation obtained Lichrospher RP-18 (250 mm × 4.6 5 μm) column isocratic mode mobile methanol 5mM sodium dihydrogen phosphate buffer pH 2.5 (60:40, v/v). flow rate maintained at 1.2 mL/min performed detector set 417 nm (λemission) after excitation 254 (λexcitation). third validation according ICH guidelines. valid concentration range 175 – 325 ng/mL AML 250 1750 OLM. proposed successfully applied analysis binary mixture two drugs formulation.
